TI New fast low-cost method of HIV diagnostics based on carbon-conjugated
antigens.

AB OBJECTIVE: To develop the new cost-effective appropriate technology of
HIV antibody detection. METHODS: Fragments of HIV genome have been
obtained by PCR from HIV-1 genome. HIV-recombinant proteins (containing
parts of gag and env) were expressed in E. coli pMal vectors, purified
and attached to the carbon particles. Recombinant proteins were absorbed
onto nitrocellulose membrane, undiluted sera were spotted and after
washing carbon-HIV proteins conjugates were added. HIV antibody were
indicated by black spots. RESULTS: Method was evaluated with
representative collection of HIV positive (130 specimens) and HIV
negative (600 specimens) sera. The sensitivity was 100%; the specificity
was 99,5%. Total assay time is approximately 10 min. DISCUSSION AND
CONCLUSIONS: Method provides simple easy to read, low-cost,
high-sensitive and specific HIV antibody detection. This technique can
be used effectively for non-instrumental diagnostics in developing
countries.
